Objectives
A mastitis-screening tool for the detection of intramammary infections with contagious mastitis pathogens will be developed for dairy herds using automatic milking system (AMS). As inflammation or an increase of somatic cell count is frequently missed in subclinical infections, detection of the infectious agent itself is the only reliable method for early diagnosis. To establish an early warning system, the use of PCR methods will be tested and samples suitable for the systematic screening of cows with a higher mastitis risk in AMS herds identified. Also the use of colostrum will be tested by bacterial cultivation.
Objectives
Für Herden mit Automatischen Melksystem (AMS) sollte ein Mastitisscreening zur Früherkennung von Infektionen mit kuhassoziierten Erregern entwickelt werden. Der direkte Erregernachweis ist dafür häufig die einzige Möglichkeit, da Entzündungsanzeichen oder ein Anstieg der Zellzahl bei subklinischen Euterinfektionen oft nicht vorliegen. Als Fragestellungen wurden untersucht, wie die PCR-Untersuchung für das Frühwarnsystem eingesetzt werden kann und welche Proben für die bakteriologische Untersuchung von Risikotieren in AMS-Betrieben genutzt werden können. Es wurde geprüft, ob sich dafür auch Proben von Erstkolostrum eignen.
Activities
For the development of the early warning system, PCR-techniques were evaluated for detection of different mastitis pathogens in milk pools that include various numbers of individual samples. In this process, the variable of interest is the optimal pool size in relation to the ct-value for each bacterial species. The aim is to establish a threshold for early indication of increasing infection rates. During the last stage of the project, the early warning system and the monitoring of cows at high risk for mastitis will be established in AMS herds to improve their udder health.
Activities
Zur Etablierung des Frühwarnsystems wurde die PCR-Untersuchung an den für die Milchleistungsprüfung automatisiert entnommenen Proben erprobt. Für jede Erregerart war dabei die optimale Poolgröße in Relation zum ct-Wert der PCR die zu bestimmende Variable. Ziel war es, einen praxisrelevanten Schwellenwert zu etablieren, der eine Zunahme der Infektionen rechtzeitig anzeigt. In der letzten Phase des Projekts wurde die Früherkennung mittels PCR sowie die Untersuchung von Risikotieren in Herden mit AMS etabliert, um deren Eutergesundheit zu verbessern.
